    Job Description

    We are seeking a highly motivated and detail-oriented Senior Research Scientist with a strong background in cell and molecular biology to join our multidisciplinary team investigating the molecular and cellular mechanisms regulating human megakaryopoiesis and hematopoietic stem cell biology.

     

    The ideal candidate will have extensive hands-on experience with primary ex vivo culture of human hematopoietic stem and progenitor cells, expertise in multicolor flow cytometry Attune NxT, molecular biology and genetic manipulation techniques.

     

    The position offers the opportunity to contribute directly to cutting-edge research aimed at understanding platelet production, stem cell regulation, and translational approaches in hematology.

    Qualifications

    + Ph.D in sciences or related field

    + Minimum five years of experience with scientific investigation.

    + Must have demonstrated outstanding achievement in a field of research

    + Must have contributed to a number of peer-reviewed publications, inventions or the like, at least some of which have had a major impact on advancing the field or discipline.

    + Prior experience successfully securing external grant funding is highly desirable.

    + Preferred:

    + 4–5 years of hands-on laboratory experience in cell biology, preferably with hematopoietic stem cells or primary human cell systems

    + Demonstrated outstanding achievement in a field of research

    + Proven expertise in flow cytometry platforms, including instrument setup, compensation design and analysis (e.g., using FlowJo, FCS Express, or similar software) of complex multicolor panels (≥8 colors).

    + Strong technical skills in protein and nucleic acid analyses (Western blot, qPCR, ELISA, etc.).

    + Demonstrated experience in human hematopoietic stem/progenitor cell isolation, culture, and megakaryocyte-erythroid differentiation including immortalized cell lines.

    + Familiarity with cell signaling pathways, cytokine stimulation assays, and phospho-protein detection.

    + Background in gene silencing (siRNA/shRNA), CRISPR/Cas9 genome editing, lentiviral transduction, and molecular cloning techniques.

    + Excellent analytical, data organization, and troubleshooting skills.

    + Ability to work independently while contributing to a collaborative team environment.

    + Must be able to work as a team member and accept constructive criticism to improve performance.

    + Prior experience studying human megakaryopoiesis, platelet biogenesis, or hematopoietic stem cell signaling.

    + Experience with colony-forming assays (CFU, CFU-Mk) and liquid culture systems for hematopoietic progenitors.

    + Experience with cell signaling pathway analysis (e.g., phospho-flow cytometry, and cytokine-inhibitor based stimulation systems).

    + Experience with gene silencing (siRNA/shRNA) or lentiviral /retroviral transduction, CRISPR/Cas9 editing.

    + Knowledge of immunofluorescence microscopy and image-based analysis for cellular morphology and megakaryocyte maturation assessment.

    + Knowledge of bioinformatics tools for flow cytometry and gene expression analysis (e.g., FlowJo, FCS Express, Prism, etc.).

    + Previous experience in translational hematology or stem cell therapy–related research.

    Responsibilities

    + Perform isolation and culture of primary human hematopoietic stem and progenitor cells (CD34⁺) and other defined subsets from mobilized peripheral blood or bone marrow.

    + Execute ex vivo differentiation assays for megakaryocyte and erythroid lineages, including morphological, molecular, and functional characterization.

    + Operate, maintain, and calibrate Attune flow cytometer, including setup-optimize of multicolor panel design for complex immunophenotyping, cell sorting and quantification of hematopoietic subsets including mature megakaryocytes and platelets.

    + Perform methyl cellulose semisolid colony-forming assays (CFU, CFU-Mk) and liquid culture systems for hematopoietic progenitors.

    + Perform protein-based assays, including protein extraction, Western blotting, ELISA, for pathway and signaling studies.

    + Conduct RNA and DNA extractions, RT-qPCR, and basic molecular biology techniques for gene expression and validation studies.

    + Perform siRNA/shRNA, CRISPR/Cas9-mediated gene editing in primary and immortalized cell systems.

    + Maintain accurate and organized experimental records, laboratory notebooks, and sample inventories.

    + Assist in the development and optimization of new protocols for cell signaling, differentiation, and functional assays.

    + Assist with data analysis, figure preparation, and contribute to manuscripts and grant applications.

    Compensation Statement

    Mount Sinai Health System (MSHS) provides a salary range to comply with the New York City Law on Salary Transparency in Job Advertisements. The salary range for the role is $58661 - $150000 Annually. Actual salaries depend on a variety of factors, including experience, education, and hospital need. The salary range or contractual rate listed does not include bonuses/incentive, differential pay or other forms of compensation or benefits.
