The Division of Cardiovascular Medicineis a dynamic and innovative center dedicated to excellence in research, medical education, and clinical care. The Division of Cardiovascular Medicine inthe Department of Medicineseeks a Life Science Research Professional (LSRP) 2 forthe Quertermous Labin the Falk Cardiovascular Research Center.
The Quertermous laboratory employs genomic and genetic methods, such as HiChIP, ATAC-seq, and ChIP-seq to identify and annotate epigenetic mechanisms upstream of causal regulatory variations that we and our collaborators have identified in large scale human genome wide association studies. In addition, our interest in CAD-associated transcription factors that are active in the blood vessel wall has led us to integrate single cell technologies into our studies. The depth of data that single cell technologies has provided allows us to characterize the gene regulatory program mediated by these transcription factors as well as the transcriptional mechanisms by which these factors modulate cell state changes.
The Quertermous lab is seeking a highly motivated LSRP to play a central role in the development and implementation of an in vivo perturb sequencing project. The goal of this project is to study CAD-associated transcription factors involved in the process of atherosclerosis by inhibiting their function in smooth muscle cells of the blood vessel wall in a mouse model of atherosclerosis. The successful candidate will have experience in the study of transcriptional regulatory mechanisms and cell biology and interest in cardiovascular disease. Interest in and experience with genetic and genomics computational methods is highly desirable.
The LSRP will perform day-to-day experiments most often utilizing mouse genetic models, cell culture, and molecular biology techniques to accomplish the goals of the lab. This work will require the maintenance of a mouse colony representing a variety of genetic mutations. S/he is expected to have some expertise in dissection of mouse, frozen sample preparation and sectioning for histology. S/he will be expected to collect, analyze, and present research data to the research group. The LSRP will be expected to troubleshoot existing protocols and develop novel protocols and procedures and provide technical and scientific advice to other research personnel in lab. Duties will also include occasional preparation of DNA, RNA, and proteins from human and animal samples and downstream assays including qPCR, Western blots, and ChIP assays. S/he will have excellent communication and organizational skills. S/he must be able to work well independently and within a diverse team.
This is a twenty-four-month fixed-term position.
Duties include:
+ Independently conduct a portion of the research project(s). Make recommendations on experimental design and/or research direction to principal investigator.
+ Independently conduct complex (lab and/or field) experiments to gather data for a portion of a major research project.
+ Interpret and analyze results, often where few guidelines exist, and make modifications to existing procedures and protocols.
+ Develop new research protocols where appropriate protocols are not described in the literature or where modification or adaptation of standard procedures and protocols is required.
+ Participate in multidisciplinary teams across different faculties or schools.
+ Perform ongoing literature review to remain current with new procedures and related research; apply to ongoing research and development of new protocols.
+ Contribute substantively to the preparation of papers for publication, especially in the results section, and to publication of findings. Present ongoing work to colleagues and/or academic conferences.
DESIRED QUALIFICATIONS:
+ Doctoral degree (PhD, MD) in related scientific field
+ 3 years of relevant work experience
+ Proficiency with molecular biology techniques, such as luciferase reporter assays, western blot, immunofluorescence, real time PCR
+ Experience with mammalian tissue culture techniques
+ Experience with in vivo techniques, such as small animal surgery, tissue sampling and histological stains
+ Experience with CRISPR techniques
EDUCATION & EXPERIENCE (REQUIRED):
+ Bachelor's degree in related scientific field and two years of relevant work experience; or Master’s degree in a related scientific field.
KNOWLEDGE, SKILLS AND ABILITIES (REQUIRED):
+ Comprehensive understanding of scientific principles.
+ General computer skills and ability to quickly learn and master computer programs, databases, and scientific applications.
+ Strong analytical skills and excellent judgment.

+ Excellent organizational skills and demonstrated ability to accurately complete detailed work.

PHYSICAL REQUIREMENTS*:
+ Frequently stand, walk, twist, bend, stoop, squat, grasp lightly, use fine manipulation, grasp forcefully, perform desk-based computer tasks, use telephone, write by hand, lift, carry, push and pull objects weighing over 40 pounds.
+ Occasionally sit, kneel, crawl, reach and work above shoulders, sort and file paperwork or parts.
+ Rarely climb, scrub, sweep, mop, chop and mix or operate hand and foot controls.
+ Must have correctible vision to perform duties of the job.
+ Ability to bend, squat, kneel, stand, reach above shoulder level, and move on hard surfaces for up to eight hours.
+ Ability to lift heavy objects weighing up to 50 pounds.
+ Ability to work in a dusty, dirty, and odorous environment.
+ Position may require repetitive motion.
WORKING CONDITIONS:
+ May require working in close proximity to blood borne pathogens.
+ May require work in an environment where animals are used for teaching and research.
+ Position may at times require the employee to work with or be in areas where hazardous materials and/or infectious diseases are present.
+ Employee must perform tasks that require the use of personal protective equipment, such as safety glasses and shoes, protective clothing and gloves, and possibly a respirator.
+ May require extended or unusual work hours based on research requirements and business needs.

The expected pay range for this position is $81,130 to $94,895 per annum.
