Responsibilities:
GENERAL PURPOSE
Primarily responsible for generating cDNA constructs encoding inositol, 1,4,5-trisphosphate receptors. This will include generating cDNAs encoding variants associated with human disease, epitope tagged constructs and concatenated constructs. The selected candidate will also be responsible for generating stable cell lines expressing these constructs as well as performing CRISPER/Cas9 experiments to engineer cell lines with mutated proteins encoded at their endogenous genetic loci. Will perform biochemical experiments to analyze expression and localization of proteins. Will perform experiments to isolate and purify protein. Will perform imaging and electrophysiological experiments to analyze the functional consequences of mutations, collect and analyze data. Will assist with overall operation of the laboratory including ordering supplies, equipment maintenance, and training of lab personnel, will help to manage projects and publish papers, and assist with preparing and submitting grants. Ability to work in a lab team environment with a diverse group of personnel is essential.
ESSENTIAL FUNCTIONS
Molecular Biology:
+ Must be proficient in RNA/DNA extraction, RT-qPCR, site directed mutagenesis, transfection and transduction.
+ Propagating plasmids. Must be proficient in CRISPR technology to generate Knock out/in genomic variants.
Protein Biochemistry:
+ Must be proficient in protein isolation, protein labelling, SDS PAGE, Native PAGE, immunopurification, Immunoblotting, Immunofluorescence and maintenance of cells in culture.
Ca2+ imaging:
+ Work in an independent manner with minimal supervision.
+ Utilize cultured cells expressing IP3R constructs for Ca2+ imaging experiments.
+ BE willing to optimize protocols for different constructs dependent on functional phenotype.
+ Must be adaptable to experimental needs.
+ Willing to appropriately analyze and interpret data in preparation for presentations and publications.
+ Must be willing to keep current with advances in. concepts and technologies.
Training responsibility:
+ Helps to interview and train laboratory personnel and rotating graduate student on laboratory and departmental-shared equipment and laboratory procedures.
+ May supervise and evaluate the work of other laboratory personnel.
+ Counsels' laboratory personnel on technical issues, problem solving, or as issues arise.
+ Develop and install operating procedures for the laboratory, operate complex equipment.
+ Perform routine equipment maintenance and troubleshooting
Electrophysiology:
+ Work in an independent manner with minimal supervision.
+ Perform voltage- and current-clamp patch clamp electrophysiology experiments.
+ Be willing to optimize protocols for different types of cells and ion channels.
+ Must be adaptable to different electrophysiological needs.
+ Must be willing to appropriately analyze and interpret data and prepare it for presentations and publications.
Research analysis, Presentation and Searches.
+ Fluent in GraphPad Prizm/Excel or similar.
+ Prepare literature searches as needed.
+ Analyze and present findings to PI and at laboratory meetings
Other job duties as assigned.
MINIMUM EDUCATION & EXPERIENCE
+ PhD in Physiology, Biochemistry, Biotechnology, or a related field Required
+ 2 years of postdoctoral training, or equivalent combination of education and experience Required
KNOWLEDGE, SKILLS AND ABILITIES
+ Required technical skills included advanced molecular biology techniques, protein biochemistry, Ca2+ imaging and some experience with electrophysiology Required
