    Qualifications

    Educational and other Requirements for the position:

    + PhD in cancer biology, hematology, immunology, or a related biomedical discipline

    + Demonstrated experience in molecular biology and murine models

    + Strong communication skills and ability to work independently and collaboratively in a team

    Experience Required:

    + Prior experience with CRISPR-based genome editing and/or xenograft models

    + Familiarity with hematopoietic stem cell biology and immune phenotyping preferred

    + Experience analyzing high-dimensional datasets (e.g., scRNA-seq) is an asset but not required

    Goals/Outcomes of the Research Project:

    + Define the molecular and cellular mechanisms by which mutant clonal hematopoiesis alters the bone marrow niche

    + Identify specific cytokines and cell types responsible for metastasis promotion

    + Evaluate targeted interventions in preclinical models

    + Generate high-impact publications and lay the foundation for future host-targeted metastasis prevention strategies

    Responsibilities

    Details of Research Project:

    The Wagenblast Lab investigates how somatic evolution in the hematopoietic system influences cancer progression. The postdoctoral fellow will focus on a project exploring how TET2-mutant clonal hematopoiesis remodels the bone marrow into a metastasis-permissive niche for solid tumors, particularly breast cancer. Using humanized xenograft models, the lab studies the inflammatory and stromal alterations induced by clonal hematopoiesis and evaluates therapeutic strategies targeting cytokine signaling. The fellow will leverage cutting-edge tools including CRISPR/Cas9 genome editing, single-cell and spatial transcriptomics, and preclinical mouse modeling to dissect how mutant immune cells orchestrate metastatic progression.

    Technical Duties: (include any protocols)

    + Generate genome-edited hematopoietic stem/progenitor cells (HSPCs) using CRISPR/Cas9

    + Perform xenotransplantation of human HSPCs into immunodeficient mice

    + Analyze bone marrow and tumor samples via flow cytometry, immunohistochemistry, and histology

    + Conduct bioluminescence imaging to monitor metastasis progression

    + Assist in spatial transcriptomics using 10x Visium HD and single-cell RNA sequencing workflows

    + Maintain primary cell cultures, perform viral transduction, and cytokine assays

    + Participate in manuscript preparation and presentation of findings

    Compensation Statement

    Mount Sinai Health System (MSHS) provides a salary range to comply with the New York City Law on Salary Transparency in Job Advertisements. The salary range for the role is $72500 - $80000 Annually. Actual salaries depend on a variety of factors, including experience, education, and hospital need. The salary range or contractual rate listed does not include bonuses/incentive, differential pay or other forms of compensation or benefits.
